Rapid iterative enzyme engineering

The drug molecules sometimes have chiral structure and are difficult to purify.

The drug molecules sometimes have chiral structure and are difficult to purify. Bota Bio can replace the conventional chemical method with a milder, safer and greener biological production approach. This transition effectively boosts yields, reduces impurities, reduces environmental pollution, and lowers production costs.

Challenges

The target product relies heavily on chemical synthesis and it has chiral structure. This leads to various by-products, making purification challenging and causing significant environmental pollution.

Solutions

We developed a machine learning model by combining public protein databases with our internal database to predict hot mutation sites. Through iterative mutagenesis, including single-site-saturation and multi-site-combination mutagenesis, and high throughput screening, we successfully improved the target enzyme’s performance.

times
With five iterative rounds of mutagenesis, enzyme catalytic activity increased nearly 900 times.
Biocatalysis has mild reaction conditions, a simplified process, and robust chiral selectivity. It is environmentally friendly, minimizes pollution and reduces costs.
